He is well-known for his roles as Charlie in Red Band Society, Mason Warner in Private Practice, and Danny Gannon in Back in the Game on television. He also played the lead in the Hulu film Big Time Adolescence. He played Sam Ecklund in both seasons of the Netflix mockumentary series American Vandal and Gabe in Locke & Key. He played Rafe Khatchadorian in Middle School: The Worst Years of My Life and Jack Dunkleman in Netflix's original film Tall Girl.
Gluck was born in Los Angeles as a child. Cellin Gluck, his father, is a film director and producer, and Karin Beck, his mother, was a production assistant and line producer. Griffin's father was born in the Japanese prefecture of Wakayama to American parents and raised in Kobe. Griffin's paternal grandparents were Japanese American Sumi (Hiramoto) and Jewish American archaeologist, historian, and Japanophile Jay Gluck.

Gluck began his acting career when he went to a summer children's showcase of Guys and Dolls at the Palisades Playhouse with his older sister, Caroline. His first significant role was as a three-year-old in Time Out, a short film co-produced by his father and directed by Robbie Chafitz.
His big break came in 2011, when he was nominated for a Young Artist Award for his performance as Michael in the film Just Go with It. He was later cast as Mason Warner on Private Practice and was later promoted to series regular. After the show was canceled, he was cast in a 20th Century Fox TV pilot called Back in the Game. It was chosen for a series. The series was canceled in November of 2013.

Gluck co-starred as Charlie on the Fox series Red Band Society in 2014, where his character, who is in a coma, serves as the show's narrator. He was cast in an NBC pilot, Cuckoo, in March 2015, but it was not picked up for a series.
Gluck made his film debut as Rafe Khatchadorian in Middle School: The Worst Years of My Life, based on James Patterson's best-selling novel of the same name.
