In 2005, she was cast as the lead in the Bollywood film Dil Jo Bhi Kahey.. (or "Whatever the Heart Says"). The film did poorly at the box office. She appeared in True True Lie (2006) alongside Jaime King and Body of Lies (2008) as Mark Strong's girlfriend. Steel Trap, a B-movie in which she starred (2008).

Her breakthrough role on television came in season three of the Showtime drama series The Tudors (2009) as Jane Seymour. She took over for actress Anita Briem. In the years since, she has appeared in the television film The Lost Future (2010) alongside Sam Claflin, as well as the ABC series Pan Am (2011). Due to a drop in viewership, the show was cancelled after one season.

Madonna directed her in W.E. : Wallis & Edouard in 2011, and she had a small role in X-Men: First Class. She was uncredited in Jared Leto's documentary Artifact and Snow White and the Huntsman in 2012.

Peaky Blinders and blockbusters from 2013 to 2017

From 2013 to 2016, Wallis played Grace Burgess in the BBC drama Peaky Blinders. Her role gave her the opportunity to be "believable in a strong, female, alpha role." The series received both critical and popular acclaim.

She joined The Conjuring Universe in 2014 as a lead in the horror film Annabelle, directed by John R. Leonetti and produced by James Wan. The film was a box office success, but it received mixed reviews. Wallis' performance was praised by critics, and she was nominated for Best Scared-As-S**t Performance at the 2015 MTV Movie & TV Awards. In Annabelle: Creation, she reappeared as Mia Form (2017).

In 2014, she co-starred with Dominic Cooper in the miniseries Fleming: The Man Who Would Be Bond as Muriel Wright, the original Bond girl.

Wallis performed nearly all of her stunts herself, including a Zero-G scene that nearly killed her. The film received poor reviews and was a box office failure. She joined Guy Ritchie's King Arthur: Legend of the Sword the same year.

2018–present: Diversification and leadership roles

In 2018, Wallis was cast as Rebecca Crosby in the American comedy Tag. The series received generally positive reviews and was nominated for Best Limited Series or Television Film at the Golden Globes. Wallis's performance was praised and seen as a "heartbreakingly powerful work" according to the Chicago Sun Times.
Later that year, Wallis returned to Peaky Blinders for the fifth season as Grace Shelby.

She appeared in Robin Pront's thriller The Silencing in 2020. The film was scheduled to premiere at the South by Southwest festival in March 2020, but it was postponed due to the COVID-19 pandemic and was eventually released on DirecTV on July 16, 2020.

Wallis also appeared as Princess Buttercup in The Princess Bride for Quibi to raise funds for World Central Kitchen in the same year, and she joined the cast of Floria Sigismondi's third film, The Silence of Mercy. The filming was completed in March 2021.
